pAUR123?DNA來源于pAUR112,是一種蛋白質(zhì)表達(dá)載體。能夠在釀酒酵母細(xì)胞中自主復(fù)制。蛋白質(zhì)表達(dá)用的啟動子是ADH1基因?(Alcohol?dehydrogenase?1?gene)的啟動子。本載體在大腸桿菌中的選擇標(biāo)記是氨芐青霉素,在釀酒酵母中的選擇標(biāo)記是Aureobasidin?A?(AbA),其抗性基因是AUR1-C,含有可以在釀酒酵母中自主復(fù)制的序列CEN/ARS。當(dāng)要表達(dá)的基因含有起始密碼子ATG時(shí),可以插入到多克隆位點(diǎn)中,能夠在酵母細(xì)胞中獲得表達(dá)。多克隆位點(diǎn)上含有三個(gè)終止密碼子,分別終止三個(gè)基因讀碼框。
