psiCHECK? -1 和psiCHECK? -2 載體用于對RNA干擾(RNAi)的早期優(yōu)化提供快速的定量方法。此載體可監(jiān)測與報(bào)告基因融合的目的基因表達(dá)的變化。兩種載體都應(yīng)用海腎螢光素酶作為主要報(bào)告基因,目的基因被克隆至海腎螢光素酶翻譯終止密碼子下游的多克隆位點(diǎn)。由合成的siRNA 或體內(nèi)表達(dá)的shRNA 引發(fā)的針對目的基因的RNAi過程,導(dǎo)致融合mRNA 的剪切和隨后的降解。檢測海腎螢光素酶活性的降低是一種監(jiān)測RNAi 效果的簡便方法。與其它的融合蛋白檢測法( 如GFP 或flag 標(biāo)簽) 相比,海腎螢光素酶方法靈敏度更高、更方便、定量更快速。推薦psiCHECK? -1 載體用于檢測活細(xì)胞中的RNAi 效果。使用EnduRen? 活細(xì)胞底物(Cat.# E6481) 檢測海腎螢光素酶活性變化,這種底物可持續(xù)監(jiān)測細(xì)胞內(nèi)的海腎螢光素酶的發(fā)光。psiCHECK? -2 載體含有第二個(gè)報(bào)告基因——螢火蟲螢光素酶, 被設(shè)計(jì)用于終點(diǎn)裂解檢測。其作用是使海腎螢光素酶表達(dá)歸一化,可以得到強(qiáng)勁且可重復(fù)的數(shù)據(jù)。
