pMAD質(zhì)粒是大腸桿菌和一些革蘭氏陽(yáng)性菌的穿梭質(zhì)粒,包括金黃色葡萄球菌、單核細(xì)胞李斯特菌、枯草芽孢桿菌等。pMAD質(zhì)粒全長(zhǎng)9666bp,含有革蘭氏陽(yáng)性菌的溫度敏感型復(fù)制起始位點(diǎn)、氨芐青霉素抗性基因和紅霉素抗性基因(erm),并含有一個(gè)耐熱的β-半乳糖糖苷酶基因(bgaB)。
pMAD質(zhì)粒可用于對(duì)一些革蘭氏陽(yáng)性菌的基因進(jìn)行失活突變。在含X-Gal的平板上,可通過(guò)藍(lán)白斑的方法快速篩選質(zhì)粒丟失菌株。在大腸桿菌中,pMAD質(zhì)粒呈多拷貝,氨芐青霉素抗性表型。在葡萄球菌等革蘭氏陽(yáng)性菌中,pMAD質(zhì)粒呈溫度敏感型復(fù)制起始,紅霉素(erythromycin)或林可霉素(lincomycin)抗性表型。
