pCDNA6.2/EmGFP-Bsd/V5-DEST實(shí)際上是一個雙表達(dá)框載體,PGK啟動子帶一個EmGFP熒光標(biāo)記基因,CMV啟動子調(diào)控外源基因的表達(dá)。不過,外源基因插入CMV啟動子后,只能用LR重組的方法實(shí)現(xiàn),所以,外源基因需要先構(gòu)建至Entry載體上,例如pENTR/D-TOPO等。
pCDNA6.2/EmGFP-Bsd/V5-DEST空載上,CMV啟動子后有ccdB表達(dá)框,所以此載體只能在DB3.1等專用的大腸桿菌中才能復(fù)制擴(kuò)增,使用此載體時,請注意配套相應(yīng)的DB3.1感受態(tài)。當(dāng)外源基因成功插入,ccdB表達(dá)框?qū)⒈恢脫Q掉,此時,重組質(zhì)粒可以在DH5α等常規(guī)大腸桿菌中復(fù)制擴(kuò)增。
