pCold-SUMO2原核表達(dá)載體是在 pCold-SUMO1載體基礎(chǔ)上將step II(TGGAGCCACCCGCAGTTCGAAAAA)標(biāo)簽加在Xba I和終止密碼子之間改造而成,該載體啟動子(CS Promoter)來源于南極嗜冷細(xì)菌,在低溫下(15 度)才能啟動蛋白的表達(dá)。低溫下細(xì)菌生長緩慢,使得蛋白合成速度減慢,從而最大限度的提高了蛋白正確折疊的幾率,提高了蛋白可溶性表達(dá),增強(qiáng)了活性蛋白的表達(dá)比率。該表達(dá)系統(tǒng)所包含的 SUMO tag可以極大地提高小分子量蛋白的表達(dá)量,而且進(jìn)一步提高了蛋白的可溶表達(dá)幾率。同時,試劑盒配備的SUMO Protease(含6×His標(biāo)簽)可特異性去除 SUMO tag,從而得到不含任何標(biāo)簽的重組蛋白。TEE信號肽可增強(qiáng)冷啟動子調(diào)控下目的蛋白的高表達(dá)。BL21(DE3)Chaprone E.coli 細(xì)菌中含有分子伴侶蛋白質(zhì)粒,其表達(dá)產(chǎn)物可協(xié)助重組蛋白正確折疊形成可溶活性蛋白。
分子伴侶蛋白質(zhì)粒為氯霉素抗性(chloramphenicol,Cmr)的表達(dá)受控于四環(huán)素(tetR)操縱子。
